MassARRAY System for medium to high throughput SNP detection

How the SNP Genotyping Works

The SNP Genotyping assay consists of an initial locus-specific PCR reaction, followed by single base extension using mass-modified dideoxynucleotide terminators of an oligonucleotide primer which anneals immediately upstream of the polymorphic site of interest. Using MALDI-TOF mass spectrometry, the distinct mass of the extended primer identifies the SNP allele.

Sample Requirements

For each SNP-plex reaction, we need 5-10µl of 50-100ng/µl genomic DNA if quantitated on nanodrop, 25-50ng/uL if quantitated with PicoGreen dsDNA quantitation assay. A260/A280 ratio should be between 1.7 and 2.0.
